The lamina explants excised from in vitro raised shoots induced multiple shoots through callus phase on Murashige and Skoog (MS) medium supplemented with cytokinins alone and in combinations. Amongst all the cytokinins namely, N6-benzyladenine (BA), kinetin (Kn), N6-(2-isopentenyl) adenine (2iP) and zeatin tried in various concentrations (0.1, 1, 5, 10, 20, 30 �M), 10 �M Kn proved optimum for differentiating an average of 15.25 � 4.72 shoots in 75% cultures within 6 weeks. Excised shoots rooted best in half strength MS adjuvanted with 1 �M indole-3-butyric acid (IBA) within 15 days of transfer. Nearly, 95.83% shoots organised an average of 6.33 � 0.67 roots per shoot. The tissue culture derived plantlets have been successfully transferred to field. Total protein analysis of leaves from in vivo, in vitro grown plants and shoot differentiating callus was carried out using SDS-PAGE, where an additional 75 kD protein band has been identified in the protein profiles of leaves from in vitro plant and this band was absent in leaves of in vivo plant.
